Principle
ChIP-Seq combines chromatin immunoprecipitation with high-throughput sequencing. A target-specific antibody enriches associated DNA fragments, enabling genome-wide studies of histone modifications, transcription-factor binding and nucleosome-related biology.
Applications
- Study histone modification patterns and their relationship to expression
- Identify binding regions of transcription factors and chromatin proteins
- Compare protein-DNA interactions between conditions
Method features
- Identifies enriched regions genome-wide
- Antibodies and sample conditions are assessed during project design
- Supports integration with ATAC-Seq, RNA-Seq and Hi-C
Workflow
- 01
Define the target protein, antibody and controls
- 02
Prepare chromatin and perform immunoenrichment
- 03
Purify DNA, prepare libraries and sequence
- 04
Identify, annotate and compare enriched regions
This is a workflow overview. Experimental and analytical details depend on sample conditions and research objectives.
