Principle
CUT&Tag uses Protein A/G-Tn5 fusion enzymes directed by antibodies to cut DNA and insert adapters near a target protein. Amplified libraries support histone modification, RNA polymerase and other compatible chromatin-target studies.
Applications
- Map histone modifications across the genome
- Investigate RNA polymerase or other compatible targets
- Prioritize candidate regulatory genes through bioinformatics analysis
Method features
- Reduces conventional immunoprecipitation and sonication steps
- Suitable for evaluating low-input and precious-sample projects
- Antibody and target compatibility are established during planning
Workflow
- 01
Prepare cells or nuclei and assess antibodies
- 02
Guide pA/pG-Tn5 to target regions using antibodies
- 03
Perform in situ tagmentation and library amplification
- 04
Sequence and evaluate target signals and enriched regions
This is a workflow overview. Experimental and analytical details depend on sample conditions and research objectives.
